Obesity is a common health concern in dogs. While factors like diet, exercise, and lifestyle all play important roles in a dog’s weight, genetics also have a significant influence.
A mutation in a gene called DENN Domain Containing 1B (DENND1B) seems to play a role as risk factor. This gene plays an important role in a brain signaling system that helps regulate appetite and energy balance (known as MC4R). When this system is disrupted, it can lead to increased hunger and reduced energy use, both of which promote weight gain.

Caractéristiques cliniques

Dogs with one DENND1B mutation show only a modest increases in food motivation and tendency to overeat while dogs with two DENND1B mutations often display clear increased appetite and a strong food-seeking behaviour, which can lead to overeating.

On average each copy of the risk allele in DENND1B can be linked to about 7–8% higher body fat in Labradors compared with non-carriers. Though this varies a lot between dogs.

Additional Information

Obesity is not caused by DENND1B alone. Multiple genes, along with environmental factors, influence a dog’s weight.
